How to Calibrate a Pipette

The need to calibrate your pipette can become a costly issue if you are thinking of sending it out to someone else. They have their costs and of course they have to raise the price to make a profit. If you follow these steps, then you can calibrate your pipette by yourself.

Instructions

Difficulty: Moderate

Step1
Adjust the dial on your pipette to the highest volume. Pipette deionized water into a flask on an analytical balance. Weigh and record the volume. Repeat five times.
Step2
Change the dial to the lowest volume. Pipette deionized water into a flask on an analytical balance. Weigh and record the volume. Repeat five times.
Step3
Change the dial to a middle range. Pipette deionized water into a flask on an analytical balance. Weigh and record the volume. Repeat five times.
Step4
Decide if discrepancies in the measurements are systematic or random. If they are systematic, then loosen the screw on the calibration wheel and turn the ring either counterclockwise to increase the volume or clockwise to decrease the volume.
Step5
Improve your technique on pipetting if the discrepancies are random.
Step6
Recheck the calibrated pipette as often as necessary.

Tips & Warnings

  • Touch the tip of your pipette to the inside wall of the flask when transferring the water.
  • Make sure to check your pipette for proper drainage before calibrating it.
