Open the Skype application by clicking "Start" > "All Programs" and selecting "Skype" from the programs list.

Hold down the "Ctrl" button and click on the two contacts with whom you'd like to initiate a three-way call.

Right-click one of the highlighted contacts and select "Call the Group." This will start a three-way call among you and the other two contacts.
