How to Get Inside the Treasure of the Romulus Room in "Assassin's Creed"
In "Assassin's Creed: Brotherhood" one of the side missions involves investigating the various hideouts of a violent, ritualistic cult called the Followers Of Romulus. The Followers inhabit six subterranean lairs around Rome. You will have to search them all in order to gain access to the Treasure Room, introduced to you through a story mission. Each lair contains a key. You will need all six keys to access the master room.

Complete the main story missions until you encounter the one about halfway through the first memory sequence. Travel into the catacombs beneath the Terme di Traiano and navigate a platforming puzzle that leads you to a large shrine room in Nero's Golden Palace. There will be several cultists in this room who will attack you. Dispatch them to access the shrine and collect the treasure. The main chest contains a scroll and a key. The scroll will describe your potential reward as a suit of powerful armor.

Locate the Treasure Room on your map. It will be marked with a Yellow Wolf icon and the shrines that contain the subsequent five keys will each be marked with a black one. The shrines will only open and become available at certain points in the story. Complete some of the missions marked with an exclamation point to progress the main campaign if there are no Black Wolf icons visible on you map.

Enter each Lair Of Romulus by pressing the "Interact" button next to the arched silver doors found at each wolf icon. Search the dark corners to find it as it will usually be quite secluded. Once inside, work your way to the shrine by solving a climbing or platforming puzzle to access a blocked-off area. There may or may not be a number of guards protecting the shrine. Collect the treasure and the key from the shrine and a secret door will open, allowing you back outside.

Collect all six keys and return to the Yellow Wolf icon on your map. You should now be able to unlock the gate to the Treasure Room.
