Fibronectin is a good medium for studying cells that adhere well to plastic plates.

Fibronectin is a glycoprotein. Glycoproteins are comprised of a protein and a carbohydrate. This glycoprotein is useful in culturing cells in a serum-free environment and cells requiring anchorage. It has been shown to increase cell survival and enhance anchorage in coated plates.

  1. Diluting for Coating

    • For coating, fibronectin can be diluted to desired concentration with buffer at pH 7-9 or serum-free culture. The final solution needs to be the correct dilute for even coating of the surface. A pH of 8 or lower can result in the formation of insoluble metal hydroxides.

    Coating Protocol

    • Plates shoud be incubated at room temperature for one hour after adding the appropriate amount of diluted fibronectin. The remaining material should be aspirated after incubation. The plates should be rinsed with distilled water. The bottom of the plates should not be scrapped. Plates may be stored at 2 degrees Centigrade to 8 degrees Centigrade or used immediately.

    Fibronectin Uses

    • Fibronectin has been shown to provide good adhesion for cells that will not bind directly on to plastic. Increased adhension and growth can result when plates are coated with fibronectin.
